In general we find you often are too prompt with your conclusion, that a mutation "has to be disease causing". In the minimise part concerning figure 2:

Did you also look at the precise positioning of the amino acids? Because Pymol just uses some kind of angle and bond defininition in order to declare a part as helix or sheet. I mean it is just a question of definition, when pymol shows you beta sheets or not.

Nice and structured entry with not too many pics. :D Or at least they are small and not overwhelming!

In table 3 you have have to get rid of: "Clashes are depicted as red and green disks" That was for table 2 I guess ;)
